05/08/2025 - During the meeting, the team presented updated results of molecular dynamics analyses, with a focus on optimising simulation parameters to improve the predictivity of the interaction between biosensor peptides and JAK2 kinase. An improvement in the stability of the simulated complexes was highlighted, thanks to the introduction of new solvation conditions.
In parallel, data obtained from a new series of ELISA experiments were discussed, conducted using cell lysates prepared with a modified lysis buffer enriched with proteasomal inhibitors. This allowed the enzymatic activity of JAK2 to be better preserved, although the phosphorylation of biosensor peptides still remained consistently undetectable.
